Advancing targeted therapies for cancers driven by the PAM pathway
Celcuity is a commercial-stage biotechnology company founded to address cellular abnormalities that drive tumor growth. Our teams of scientists, biochemists and molecular biologists are working to advance new targeted therapies for cancers involving the PAM (PI3K/AKT/mTOR) pathway, a key driver of many tumor types, including breast, prostate, lung, endometrial, colorectal and ovarian cancers.
The company’s first approved product, REVTORPYK (gedatolisib), targets all Class I isoforms of PI3K and the mTOR complexes, mTORC1 and mTORC2, to comprehensively block the PAM pathway. REVTORPYK’s mechanism of action and dosing schedule are differentiated from other currently approved and investigational therapies that target PI3Kα, AKT, and/or mTORC1 alone or together.
The PI3K/AKT/mTOR, or PAM, pathway is one of the most important targets in cancer but comprehensively inhibiting it has stymied researchers and drug developers for nearly two decades. REVTORPYK addressed this 20-year challenge by becoming the first pan-PI3K, mTORC1/2 inhibitor approved by the FDA. We are thankful for the opportunity to make this important new therapy available to patients.

To expand the number of patients who may potentially benefit from REVTORPYK, we are conducting two global Phase 3 clinical trials to evaluate REVTORPYK as first-line treatment for patients with HR+/HER2- locally advanced or metastatic breast cancer and a Phase 1/2 clinical trial to evaluate REVTORPYK as second-line treatment for patients with metastatic castration resistant prostate cancer.
